Abstract

Designing bispecific antibodies is a complex process, enabling drug developers additional levels of optimization and fine-tuning of lead compounds. Though the task may be daunting, some common challenges of generating bispecific antibodies have been tackled by protein engineers with creative approaches, and several methods for determining the sequences of potential binders to incorporate into the bispecific antibodies are available. Suggested initial go-to formats will be presented and the rationale behind starting off bispecific antibody campaigns with such formats. The utility of mouse bispecific antibodies in basic research and drug discovery will also be discussed including specific examples of their use.

Protein Sequencing and Characterization to aid Design and Testing of Bispecific Antibodies

Determining the protein sequence and other characteristics of mAbs prior to bispecific engineering provides important guidance. Confirming the sequence of the protein produced is as important to discovery and engineering as it is to later production of bispecific antibodies. In this presentation, scalable methods for protein sequencing, protein sequence confirmation and characterization of antibodies and their epitopes will be discussed.
